Step 4-1 Sketch a basic P&ID for a media sterilizing tank arrangement with the following main equipment components (your P&ID sketch can be hand-drawn or done using a basic software drawing package): • Media sterilizing tank with heating and cooling jacket • Bottom mounted agitator • Inline process air filter • Inline process liquid filter • Inline exhaust filter
Include the following process and utility connections on your media sterilizing tank P&ID 1. Unsterilized media supply into media sterilizing tank 2. Supply of heat-labile sterile constituents (small quantities) via the inline process liquid filter (post media sterilization) 3. Tank bottom outlet routed to bioreactor (with bypass to industrial waste) 4. Process air supply, via inline sterilizing filter, to media sterilizing tank (for pressure transfer of sterilized media to the bioreactor) 5. Tank vent-control exhaust line routed to a high level via the inline exhaust filter 6. CIP supply to media sterilizing tank via sprayball 7. Multiple clean steam supplies to media sterilizing tank (to sterilize tank and all process connections), and associated contaminated condensate routed to drain/waste 8. Over pressurization tank vent line to low level bund via a rupture disc 9. Plant steam supply and condensate return from the tank’s heating and cooling jacket 10. Plant cooling tower water supply and return from the tank’s heating and cooling jacket 11. Non-sterile process-air supply to ‘blow-down’ tank’s heating and cooling jacket
